dc.description.abstract | Spracovateľ: Lenka Žiaková Názov diplomovej práce: Izolácia alkaloidov z Narcissus poeticus recurvus a ich biologická aktivita IV Univerzita: Univerzita Karlova, Farmaceutická fakulta v Hradci Králová, Katedra farmakognózie a farmaceutickej botaniky Školiteľ: PharmDr. Rudolf Vrabec, Ph.D. Oponent: PharmDr. Marcela Šafratová, Ph.D. V tejto diplomovej práci boli pomocou chromatografických separačných metód spracované vopred pripravené jednotlivé frakcie z alkaloidných extraktov rastliny Narcissus poeticus recurvus. Táto rastlina patrí do čelade Amaryllidaceae, čo je čeľaď s významým zdrojom alkaloidov, ktoré inhibujú AChE/BuChE a zmierňujú tak symptómy súvisiace s Alzheimerovou chorobou. Chromatografiou - konkrétne TLC chromatografiou - boli vyizolované v čistom stave 4 alkaloidné látky (inkartín, narciklasín a dva rôzne izoméry brunsvigínu). Ich štruktúra bola overená metódami GC/MS, NMR, polarimetria a v prípade izomérov brunsvigínu aj pomocou cirkulárneho dichroizmu, kde sa však ale absolútnu konfiguráciu nepodarilo určiť. Inhibičná aktivita získaných látok bola hodnotená voči ľudským cholinesterázam (hAChE/hBuChE), ale žiadna z nich nevykazovala významný inhibičný efekt (IC50 > 100 μM). dc.description.abstract | Author: Lenka Žiaková Title of the Thesis: Isolation of Alkaloids from Narcissus poeticus recurvus and Their Biological Activity IV University: Charles University, Faculty of Pharmacy in Hradec Králové, Department of Pharmacognosy and Pharmaceutical Botany Supervisor: PharmDr. Rudolf Vrabec, Ph.D. Opponent: PharmDr. Marcela Šafratová, Ph.D. This diploma thesis focuses on the chromatographic separation methods applied to pre- prepared individual fractions from alkaloid extracts of the Narcissus poeticus recurvus plant. This plant belongs to the Amaryllidaceae family, which is a significant source of alkaloids that inhibit AChE/BuChE and thus alleviate symptoms related to Alzheimer's disease. Chromatography - specifically TLC chromatography - was used to isolate four alkaloidal compounds in pure form (incartine, narciclasine, and two different isomers of brunsvigine). Their structures were verified using GC/MS, NMR, polarimetry; in the case of brunsvigine isomers, circular dichroism was also applied, although it did not resolve their absolute configuration. The inhibitory activity of the obtained compounds was evaluated against human cholinesterases (hAChE/hBuChE), but none of them exhibited significant inhibitory effects (IC50 > 100 μM).
